Sue Ursula Bertossa Dr Kate Fairweather & Professor Sharon Lawn A phenomenological study of gambling and Vietnamese Australians. Problem gambling is a significant health problem in Australia; despite the cultural diversity of our nation little is known about the role culture plays in defining this issue. This study adopted a phenomenological approach, creating an understanding of what it means to be a problem gambler within a Vietnamese Australian context. The comparatively libertarian nature of commercial gambling in Australia has proven popular with Vietnamese Australians; venues offer a welcoming, exciting, and non-discriminatory environment, helping overcome the isolation and loneliness often associated with migration. Gambling has also provided a compelling emotional escape for those affected by trauma; and it’s sometimes perceived as a means for creating much-needed capital. Yet, Vietnamese Australians recognise the risk of becoming dependent on gambling, and identified the prominent role that shame, in all its cultural complexities, treat problem gambling.
Katherine Mary Boschen Professor Sharon Lawn & Dr Caroline Phelan NDIS Participants with Psychosocial Disabilities: Investigating their formal supports following a life-limiting diagnosis The National Disability Insurance Scheme (NDIS) has supported Australians with disabilities for almost a decade. To ensure the sustainability of the NDIS, Australian Governments have agreed that where services are best funded under universal service obligations or via another service system, such as health or mental health, the NDIS will not fund those supports. Formal support systems have not yet agreed on how they plan to assist NDIS participants once they develop life-limiting conditions, particularly those without pre-existing physical disabilities, such as participants with psychosocial disabilities. This research’s significant and original contribution to knowledge is a detailed analysis of the types of support formal systems currently provide participants with psychosocial disabilities and life-limiting conditions. The research contrasts the expectations of participants and their informal and formal supports with the intergovernmental agreements, identifies the gaps and barriers, and reveals whether the support ensures equity and seamlessness between systems.
Manam Inushi De Silva Professor Cedric Bardy, Professor Simon Conn & Professor Brett Stringer Glioblastoma plasticity and therapeutic sensitivity in the human brain microenvironment Glioblastoma (GBM) is a common, but highly aggressive brain tumour. Despite advances in treatment, it is difficult to treat because of its ability to adapt and survive in different environments. This thesis examines how GBM tumours adapt to brain environments making them resistant to chemotherapy and radiation. A major finding is that cerebrospinal fluid (CSF), which normally supports brain cells, can make GBM tumours harder to treat. The research highlights a potential solution: repurposing an antipsychotic drug called trifluoperazine (TFP). TFP blocks proteins that promote treatment resistance and makes GBM more sensitive to chemotherapy and radiation. While TFP shows promise, more research is needed on how it works in the body. This thesis also suggests that lab-grown, human brain cell models could improve GBM research. These models reflect real brain diversity, making them useful for testing treatments and understanding how GBM affects healthy brain cells.
Jose Javier Estevez Bordon Professor Jamie Craig, Professor John Landers, Associate Professor Richard Mills, Alex Brown & Dr Natasha Howard Defining the Epidemiology of Blindness and Diabetic Retinopathy among Aboriginal and Torres Strait Islander People This research delved into the escalating issue of diabetes and its dire impact on vision among Aboriginal and Torres Strait Islander people, a group facing significant health disparities. Focused on understanding the complex social, clinical, and biological risk factors underpinning diabetes-related blindness and other vision impairments, the study utilised data from landmark studies. These studies shed light on the risk and outcomes associated with diabetes. Key findings highlighted the disproportionate burden of visual disabilities within this community, influenced by modifiable factors like education and employment, and underscored by the pervasive challenge of accessing quality eye care and screenings. This research not only fills a critical gap in our understanding of diabetes-related vision loss but also underscores the urgent need for tailored, accessible healthcare solutions to mitigate these risks among Australia's Indigenous population, paving the way for a more inclusive and effective healthcare framework.
Anowarul Islam Dr Stephen Gregory, Dr Binoy Appukuttan & Professor Leanne Dibbens Metabolic effects of aneuploidy and their relevance to cancer and neurological phenotypes Chromosomal instability (CIN), a hallmark of most human solid tumors, causes ongoing chromosomal gains or losses, leading to aneuploidy. Aneuploidy contributes to cancer progression, poor prognosis, and drug resistance. Targeting aneuploidy for cancer treatment requires understanding signaling pathways enabling aneuploid cell survival. Using Drosophila as a model, I investigated transcriptional responses to CIN and identified altered one-carbon metabolism, particularly S-Adenosyl methionine (SAM) pathways. Depleting SAM synthase in CIN cells induced apoptosis, reversible by spermine supplementation. Loss of polyamines heightened ROS susceptibility and reduced autophagy, driving CIN cell death, suggesting polyamine inhibition as a therapeutic strategy. Additionally, I modeled neuronal aneuploidy in Drosophila, linking Mad2 depletion in GABAergic neurons to increased aneuploidy, cell death, and seizure phenotypes, mitigated by antioxidants. Finally, I developed Drosophila lines expressing human KCNT1 epilepsy mutations, demonstrating seizure phenotypes modifiable by cannabidiol. This establishes Drosophila as a model to study CIN-related diseases and KCNT1 epilepsy.
Andrea Jane Mangion Professor Neil Piller & Professor Sue Gordon Digital health in lymphoedema care Telehealth adoption surged during the COVID-19 pandemic, allowing continued care for lymphoedema patients during lockdowns. Before this eHealth modalities, including telehealth, were scarcely researched in lymphoedema care and the extent of the utilisation of digital health was largely unknown. Through sixteen publications from 2021 to 2024 and the development of five digital prototypes—a chatbot, online course, educational app, serious game, and digital assessment tool—this thesis significantly contributes to the understanding and application of digital health in lymphoedema care. Eighteen eHealth modalities were identified as being utilised in various aspects of prevention, diagnosis, assessment, and treatment. This work is original, significant, and timely, given the rapid expansion globally of digital health technologies beyond telehealth to include sophisticated applications of robotics and artificial intelligence.
Jai Lachlan Meyers Professor Robyn Meech & Research Fellow Julie-Ann Hulin The Role of UGT Enzymes as Novel Modulators of Lipid Biosynthesis in Breast Cancer The human UDP-glycosyltransferase (UGT) superfamily is comprised of 22 enzymes, with many having well defined functions in disease. Although previously linked to pathogenic features of breast cancers, the precise biological activities of the UGT isoforms; UGT2B11 and UGT2B28, were largely undefined. This research explored the role of these two orphan enzymes in breast cancer and attempt to deduce their functional roles. Specifically, this project examined associations of UGT2B11 and UGT2B28 expression levels with clinical and biochemical parameters in breast cancer patient cohorts. This led to defining a role of these UGTs in modulating the activation of SREBPs (sterol regulatory binding proteins), the master regulators of lipid biosynthesis. Finally, a role of these UGTs in mediating crosstalk between SREBP and androgen receptor signalling was identified. Overall, this research has demonstrated the important role of UGT2B11 and UGT2B28 as novel modulators of lipid biosynthesis in breast cancer.
Harman Sharma Dr Luke Grundy, Dr Steven Taylor & Professor Simon Brookes Role of Bladder Sensory Neurons in Urinary Tract Infection and Chronic Bladder Hypersensitivity My thesis investigates bladder's intricate nerve network and how urinary tract infections (UTIs) can disrupt its function. The bladder relies heavily on sensory and autonomic nerves to maintain proper function and UTIs can lead to alterations in these pathways, causing chronic pain and dysfunction. By using a mouse model, I found that UTIs significantly increase nerve sensitivity and alter bladder function as a result of infection-induced inflammation. Importantly, I discovered that persistent changes in nerve activity and gene expression can remain long after the infection, predisposing individuals to ongoing bladder issues. This work fills a critical knowledge gap about the neural mechanisms behind acute and chronic bladder conditions, underscoring the need for treatments that target underlying nerve dysfunction rather than just managing symptoms. By revealing how bladder nerves contribute to pain and sensitivity, this research offers important insights that could lead to better and effective therapeutic approaches.
